当归芍药散对药物性肝损伤小鼠的保护作用及其机制研究

Protective Effect of Danggui Shaoyao San on Drug-Induced Liver Injury in Mice and Its Mechanism

摘要: 观察当归芍药散对异烟肼(INH)和利福平(RFP)合用致小鼠药物性肝损伤的保护作用及其机制。将小鼠随机分为正常组、模型组、当归芍药散高、中、低剂量组(32,16,8mg/kg)和联苯双酯组(200mg/kg)。采用INH和RFP合用复制小鼠药物性肝损伤模型,测定血清中谷丙转氨酶(ALT)、谷草转氨酶(AST),同时测定肝匀浆中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)和丙二醛(MDA)含量;HE染色法观察肝组织病理学变化,分析当归芍药散的保肝作用及其可能机制。结果表明当归芍药散可降低INH和RFP联用所致药物性肝损伤小鼠肝脏系数,显著降低小鼠血清中ALT(P<0.01),降低AST(P<0.05)含量;肝脏MDA含量明显降低(P<0.05),SOD活力显著提高(P<0.05),GSH-Px含量呈增高的趋势;病理学检查表明当归芍药散能显著减轻肝损伤程度。文章指出当归芍药散对药物性肝损伤小鼠具有一定的保护作用,其机制可能与其抗氧化作用有关。

Abstract: To investigate the protective effect and mechanismof saponins of Danggui Shaoyao San(DSS)on drug-induced liver injury in mice. Method mice were randomly divided into six groups,control group,low,middle,high-dose (8,16,32mg/kg)of DSS,model group and bifendate group(200mg/kg).The model of drug-induced liver injury was induced by intragastric administrationof isoniazid(INH) and rifampin(RFP) in mice.The levels of alanine aminotransferase(ALT),aspartate transaminase(AST)in serum and the content of superoxide dismutase(SOD), malondialdehyde(MDA),glutathione peroxidase(GSH-Px)inliver homogenate were assayed by spectrophotometry. The pathological changesof hepatic tissueswere observed by hematoxylin and eosin(HE)staining.Result The liver index DSS middle,high-dose groups was lower than the model group(P<0.05 or P<0.01).Compared with the model group,the levels of ALT and AST In DSS middle,high-dose groups were significantly decreased( P<0.05 or P<0.01). The content of MDA DSS middle,high-dose groups was also lower than that in the model group(P<0.05 or P<0.01), while the activity of SOD and GSH-Px in DSS middle,high-dose groups was higher than the model group(P<0.05 or P<0.01).Hepatic tissues stained with HE showed that PNS could remark ablyre lieve liver injury.The article points out DSS has protective effect on drug-induced liver injury in mice,which may be related toantioxidant effect.
