HPLC-ELSD法测定薏仁红枣饮中甘油三油酸酯含量

Determination of Glycerol Trioleate in Hongzao Yiren Yin by HPLC-ELSD Method

摘要: 目的:采用HPLC-ELSD法对薏仁红枣饮及不同产地的薏苡仁中的甘油三油酸酯成分进行含量测定。方法:运用ALLtima C18色谱柱,流动相乙腈-二氯甲烷(55∶45)和(63∶37),流速1mL/min,蒸发光散射检测器的氮气流速为1.5L/min、漂移管温度为50℃。结果:10个不同产地的薏苡仁中甘油三油酸酯的线性范围为0.28~2.80μg,r=0.9992,平均加样回收率为99.34%,RSD=0.34%;其中甘油三油酸酯含量最高是福建莆田产薏苡仁,为0.971%。薏仁红枣饮中甘油三油酸酯的线性范围为0.28~2.80μg,r=0.9997,平均加样回收率为101.11%,RSD=1.07%,其中甘油三油酸酯平均含量为0.162%。结论:该方法操作简单方便,方法准确,重复性好。

Abstract: To determine the content of Glycerol Trioleate in Hongzao Yiyiren Yin and the different regions of Semen Coicis by HPLC-ELSD. Methods: The ALLtima C185μ columns was used with mobile phase of acetonitrile-dichloromethanol (55∶45) and (63∶37) at the flow rate of 1.0 mL/min. The 1.5L/min nitrogen flow for the evaporative light scattering detection and the tube temperature of 50℃. Results: The linear range of Glycerol Trioleate in the different area of Semen Coicis was 0.28~2.80μg, r=0.9992, the average recovery was 99.34%, RSD=0.34%. The highest Glycerol Trioleate content of Semen Coicis in ten different area from Putian, Fujian, was 0.971%. Glycerol Trioleate in the linear range of Hongzao Yiyiren Yin was 0.28~2.80μg, r=0.9997, the average recovery was 101.11%, RSD=1.07%. The Glycerol Trioleate content in Hongzao Yiren Yin was 0.162%. Conclusion: The method is simple, accurate and repeatable.
