尿素包埋法纯化DHA油脂的研究

Investigation on Purification of DHA Oil Produced by Urea Complexation

摘要: 以由裂殖壶菌(Schizochytrium sp)发酵而得的DHA精油为原料,运用尿素包埋法处理,分别分析了尿素包埋反应中包埋温度、包埋时间、尿酯比、溶酯比对混合脂肪酸乙酯中DHA乙酯富集效果的影响,根据单因素的实验结果,运用响应面法进行实验设计,考查了各因素之间的交互作用,借助响应面设计软件分析了实验结果,得到尿素包埋的最佳工艺参数为包埋温度-5℃,包埋时间6 h,尿酯比1.2,溶酯比12。在此条件下进行验证性实验,所得实验结果为DHA乙酯含量为56.62%,富集相收率为62.42%。

Abstract: Urea complexation was applied to increase the content of DHA fermented by Schizochytrium sp. The DHA was esterified with ethanol solution of KOH to acquire mixed fatty acid esters. The conditions of urea complexation in purifying DHA were optimized through single factor test and response surface method. The optimal condition was as follows:complexation time 6h, complexation temperature-5℃, urea-to-oil ratio(w/w)1.2 and ethanol-to-oil(v/w) 12. Under the optimal conditions, the DHA ester (DHA-E) content and yield of enrichment phase were 56.62%and 62.42%, respectively.
